The presence of dendritic cells, often known as “sentinel” immune cells is critical for maintaining and regulating the body’s immunological response balance. Researchers have revealed that these cells play an important role in the therapy of cancer and severe viral infections. Chronic viral infections and tumors can damage the immune system permanently, lowering the ability of immune killer T cells to destroy tumor cells or those infected with a virus – this is known as ‘immune fatigue.’ Blocking immune fatigue using medications like checkpoint inhibitors has shown to be a very effective therapy for some malignancies, however, these immunotherapies may not always work and can have serious adverse effects.

RESEARCH ON T-CELL ANTIGEN RECEPTOR (TCR)

The emergence of immunotherapeutic techniques marked a milestone in cancer therapy. Immunotherapy, specifically immune checkpoint inhibitors, bispecific antibodies, chimeric antigen receptor (CAR) T-cell therapy, and others, has been shown to significantly improve progression-free survival and overall survival in patients with a variety of malignancies, including melanoma, lung cancer, acute lymphoblastic leukemia, colorectal cancer, and others. Despite these results, the vast majority of patients who are eligible for immune treatment do not benefit from it due to intrinsic or secondary resistance to therapy.

Dr. Paul Ernest Love developed one of the first gene-targeted models at NIH as a postdoctoral fellow, mice missing expression of the CD3zeta chain of the T Cell Antigen Receptor (TCR). As a result, his lab has created mice defective in multiple different TCR subunits. Understanding the nature and function of the TCR’s numerous signal-transducing modules known as Immuno-receptor-Tyrosine-based-Activation-Motifs (ITAMs) has been a long-term focus of research at the Love-laboratory. T cells, one of the lymphocyte types required to combat infectious diseases, utilize ITAMs. T cells also identify and eliminate cancer cells and play an important part in self/non-self-identification.

Previous attempts to design T cells focused on targeting specific tumor types by the careful selection of cancer or tissue-specific proteins (antigens). Since its inception more than 10 years ago, chimeric antigen receptor (CAR)-T cell therapy has shown to be extremely efficient in targeting blood cancer cells, resulting in several FDA-approved CAR-T medicines. CAR-T cells contain antigen receptors on their surface that detect and target particular proteins found on cancer cells. Some patients are cancer-free even a decade after receiving CAR-T cell treatment, owing to the T cells implanted years before. However, one of the greatest hurdles facing biomedical research is determining why the vast majority of cancer patients who get CAR-T cells fail to achieve remission.

Dr. Paul Ernest Love and his colleagues have recently launched experiments to use their insights about human TCR signaling components and TCR signal altering proteins in tumor immunotherapy. This study, which might lead to a game-changing breakthrough in immunotherapy, focuses on the TCR’s signaling machinery, either directly or indirectly. Two Love-lab research lines have identified potential translational implications of fundamental science for human medicine. One line focuses on the structure and function of the TCR, with a particular focus on the signal-transducing ITAMs found within the TCR’s six signal transducing subunits.

Paul E Love’s group has been involved in partnerships that resulted in the creation of a mouse model for human SCID-X1, as well as animals lacking in or expressing mutant variants of the essential T cell signaling adaptor LAT or the chemokine receptor CCR9. Members of the Love-lab have also discovered key functions for the proteins CD5, CD69, Txk, Themis, Themis2, and Fbxl12 in T cell development using genetic screening methodologies and gene targeting studies. Another study from the Love lab found that multiprotein transcriptional complexes comprising the proteins Ldb1 and Lmo2 play an important role in hematopoietic stem cell maintenance/self-renewal, erythropoiesis, and the development of T cell acute lymphoblastic leukemia.
